一株高温好氧驱油菌的筛选及油藏适应性分析

摘要
胜利油田油藏温度偏高,多数在60~80℃之间,一般驱油用好氧微生物的生长温度为30~60℃,温度成为利用微生物驱油的主要限制条件,针对这一特点,对高温油井产出液中的菌种在培养基中经反复分离纯化,筛选到一株性能较好的高温好氧菌种ZJ-3,该菌生长温度、耐矿化度能力、耐酸碱性等理化性能分析及通氧量对菌种生长及代谢产物的影响试验结果表明,ZJ-3最适宜生长温度为70℃,最高生长温度达80℃,经121℃高压灭菌30 min仍能存活,其生长代谢可产生表面活性剂,可耐受溶质质量分数最高5%的矿化度,能利用原油为唯一碳源生长,适合一般高温油藏,通氧量对细菌的代谢产物影响较大。物理模拟试验表明,该菌可在一次水驱基础上进一步提高采收率达12%。该高温好氧菌种在微生物提高采收率技术中有良好应用潜力。
Abstract
Most reservoir temperature of shengli oil filed is between 60 to 800℃,but the normal aerobic microbe growth adapt in 30 to 600℃.In view of limit of temperature on microbe growth,separated and purified one of aerobic bacteria ZJ-3 from produced fluid of a hot hole,the experiment of growth temperature,salinity-resisting,Acid-alkali-resisting and metabolite shows,ZJ-3 adapt to growth in 700℃ circumstance,and the maximum temperature can reached to 800℃,and it can survive at 1200℃ and exterminate bacterium for 30 minutes. The surface acting agent metabolized by JZ-3 can resist 5 salinity solute,it is adequate to normal reservoir and growth with carbon of oil,and the oxygen has great effect on metabolize of bacteria.Physical experiment shows,ZJ-3 can enhance oil recovery to 12,and has favorable utilization foreground.
